At what location/file is documented/configured how much of a good will be placed in one box for transport ?

It is written in file goods.dat (and its pak mirror file - goods.pak or good.<name of good>.pak)

Yes, you would be looking for metric and  weight_per_unit:

obj=good
name=Buecher
mapcolor=114
metric=paletten
catg=1
value=330
speed_bonus=3
weight_per_unit=250

Simutrans doesn't really deal with boxes. Box is just a custom name for the unit for some types of goods. As such, the answers are more to the question "How do you name the smallest unit for a type of goods and how much does it weigh?"
