I've set out 2 conjoint DLR B07's and set it on a route but the back two carriages are always empty while the ones at the front take the load.

then maybe the loading station is too short, so that the train does not fit into the station.

does that affect trams as well? I forgot to mention that its a tram. Even so it should affect it as the front two carriages dont fit the station anyway and still load
How long are the stations? If you have 2 2-car B07s back-to-back, they need two tiles of station. The people can't board at the back because the train is too long for the station. (if you're not sure how many a combination of trains takes at a station, look at it in the depot window). This is the same for trams and trains.
A single 2-car B07 takes exactly one tile and therefore fits in a single station tile.
